WT1基因在NSCLC中的表达及其临床意义

摘要:

目的 探讨Wilms tumor gene 1(WT1)基因在非小细胞肺癌(non-small-cell lung cancer,NSCLC)中的表达及其临床意义。方法 通过实时荧光定量PCR技术检测100例NSCLC组织石蜡标本及对照标本(40例癌旁组织和40例良性病变组织)中WT1基因的表达量,以对照组WT1相对表达量均值为比较标准,将癌组织WT1表达量分为高表达和低表达;分析WT1表达量与NSCLC患者临床特征、生存期之间的关系。结果 与对照组组织比较,WT1基因在NSCLC组织中高表达[(4.295±1.477) vs (1.001±0.002),P<0.01]。WT1基因表达在NSCLC患者病理类型[(4.132±0.576) vs (6.118±0.525)]、分化程度[(3.973±0.329) vs (5.683±0.383)]、TNM分期[(4.829±0.137) vs (5.685±0.473)]和淋巴结转移[(3.165±0.499) vs (5.186±0.618)]方面比较,差异均具有统计学意义(均P<0.01)。WT1高表达患者(WT1表达 ≥ 1.001)生存期缩短(43个月 vs 59个月,P=0.002)。结论 WT1基因在NSCLC组织中高表达,且高表达能够促进NSCLC分化和转移,缩短患者生存期。

Objective To detect the expression and clinical significance of Wilms tumor gene (WT1) in non-small-cell lung cancer (NSCLC). Methods Quantitative PCR (qPCR) was used to detect the expression level of WT1 in 100 NSCLC tissue samples (the NSCLC group). Forty paracancerous tissue samples and 40 benign tissue samples were used as control. NSCLC samples were divided into the high expression group and the low expression group according to the comparison with the average expression of WT1 in the control group. The relationship between WT1 expression, the clinical characteristics and the survival of NSCLC patients were further analyzed. Results WT1 has a higher expression in NSCLC group than that in the control group[(4.295±1.477) vs (1.001±0.002), P<0.01]. Moreover, WT1 expression was significantly different in terms of pathological types[(4.132±0.576) vs (6.118±0.525),P<0.01], differentiation[(3.973±0.329) vs (5.683±0.383),P<0.01], TNM stage[(4.829±0.137) vs (5.685±0.473), P<0.01], and lymph node metastasis[(3.165±0.499) vs (5.186±0.618),P<0.01]. Finally, NSCLC patients with higher WT1 level (≥ 1.001) had shorter survival length than those with lower WT expression (43.0 month vs 59.0 month, P=0.002). Conclusion WT1 gene is highly expressed in NSCLC tissues, and its high expression can promote the differentiation and metastasis of NSCLC, as well as shorten the survival time of the patients.
